== Ion-exchange chromatography == DEAE-C is a weak anion exchanger. This exchange is utilized to separate proteins that have faintly differing charges. Like all anion exchangers, the resin carries a positive charge that interacts favorably with negative charges. The positive charge of DEAE cellulose is due to a protonated amine group. To ensure that the resin is protonated and positively charged, the chromatography should be performed at least 2 pH units below the pKa of the amine group, 10. The strength of the bond between the resin and protein is highly dependent on the pH range in the column and the pI of the protein of interest. The resin is a weak exchanger because it is only partially ionized over most pH values, and an efficient separation with DEAE-C chromatography requires a specific, narrow pH range. Cellulose, dextran, agarose, and other insoluble complexes are unaffected because they compose inert matrices, hence why they are so often derivatized with strong and weak cation and anion exchangers in chromatography. DEAE-C beads have diethylaminoethyl chains covalently bound to oxygen atoms on the D-glucose subunits of cellulose.
=== Related books === In 1991, Pryor published his first book about Flag Fen, entitled Flag Fen: Prehistoric Fenland Centre, as one of a series co-produced by English Heritage and B.T. Batsford. The final monograph on the site – entitled The Flag Fen Basin: Archaeology and environment of a Fenland Landscape – was published in 2001 as an English Heritage Archaeological Report. The report is now available online through the Archaeology Data Service. Pryor has followed this with a third book on the site, published by Tempus in 2005. Entitled Flag Fen: Life and Death of a Prehistoric Landscape, it is what he has described as a "major revision" of his 1991 work, for instance repudiating his earlier "lake village" concept.
== History == This cone was described by Sir Geoffrey Ingram Taylor in 1964 before electrospray was "discovered". This work followed on the work of Zeleny who photographed a cone-jet of glycerine in a strong electric field and the work of several others: Wilson and Taylor (1925), Nolan (1926) and Macky (1931). Taylor was primarily interested in the behavior of water droplets in strong electric fields, such as in thunderstorms.

In 2017, Guinness teamed up with AB InBev to distribute Guinness in mainland China. China is the single biggest worldwide alcohol market, especially for imported craft beers like Guinness. Molecular studies have revealed an unexpectedly large number of cryptic lichen species. Cryptic species are genetically distinct yet morphologically similar to other species. Because many lichens differ only subtly in form or chemistry, they harbour extensive cryptic diversity. Lumbsch and Leavitt (2011) termed this a paradigm shift: morphology alone often fails to delimit species. They reviewed numerous examples across lichenology. In the large foliose lichen genus Xanthoparmelia, eight morphospecies were merged after DNA showed they formed a single lineage. Conversely, lookalike Xanthoparmelia thalli fell into several distinct lineages, increasing species counts. Genetic work on the pantropical Cladia aggregata complex uncovered at least 11 distinct species within what had been treated as one. A phylogenomic survey of the cosmopolitan granite‑dweller Lecanora polytropa pushed this pattern to the extreme, recovering up to 75 evolutionarily independent lineages within what had long been treated as a single species—about a seventy‑fold jump in recognized diversity. Such cases show that traditional concepts both lumped and split species incorrectly. Characters like growth form, colour, or the presence/absence of sexual structures (such as whether a lichen reproduces by spores or is sterile and reproduces only asexually by fragments) were often over-weighted and do not always track evolutionary lineages.

Synthesis of RNA typically occurs in the cell nucleus and is usually catalyzed by an enzyme—RNA polymerase—using DNA as a template, a process known as transcription. Initiation of transcription begins with the binding of the enzyme to a promoter sequence in the DNA (usually found "upstream" of a gene). The DNA double helix is unwound by the helicase activity of the enzyme. The enzyme then progresses along the template strand in the 3' to 5' direction, synthesizing a complementary RNA molecule with elongation occurring in the 5' to 3' direction. The DNA sequence also dictates where termination of RNA synthesis will occur. Primary transcript RNAs are often modified by enzymes after transcription. For example, a poly(A) tail and a 5' cap are added to eukaryotic pre-mRNA and introns are removed by the spliceosome. There are also a number of RNA-dependent RNA polymerases that use RNA as their template for synthesis of a new strand of RNA. For instance, a number of RNA viruses (such as poliovirus) use this type of enzyme to replicate their genetic material. Caesium-137, along with other radioactive isotopes caesium-134, iodine-131, xenon-133, and strontium-90, were released into the environment during nearly all atmospheric nuclear weapon tests, and more recently some nuclear accidents, most notably the Chernobyl disaster, the Goiânia Accident and the Fukushima Daiichi disaster. Caesium-137 is produced from the nuclear fission of plutonium and uranium, and by observing the characteristic gamma rays emitted by this isotope, one can determine whether the contents of a given sealed container were made before or after the first atomic bomb explosion (Trinity test, 16 July 1945), which spread some of it into the atmosphere, quickly distributing trace amounts of it around the globe. This procedure has been used by researchers to check the authenticity of certain rare wines, most notably the purported "Jefferson bottles". Surface soils and sediments are also dated by measuring the activity of 137Cs.
== History == Fascism and Freedom Movement was founded in 1989 as a political movement inside the Italian Social Movement, centered around Senator Giorgio Pisanò and his weekly Candido. Finally, it separated from the MSI on 25 July 1991. In 2009, it added the words National Socialist Party (NSP) to its original name. The movement has been sued several times for alleged reconstitution of the dissolved National Fascist Party. It refuses agreements and / or alliances with other neo-fascist parties, stressing categorically not to be classified as a right-wing party, and that similar parties do not sufficiently adhere to Fascist ideology. The party makes explicit reference to the ideals of the Italian Social Republic as corporatism and the nationalization of the economy. Their ideology is based on the thought of fascist leader Benito Mussolini, and has as its main objective the realization of Mussolini's strongly hierarchical corporate democracy based on social credit. The party is also in favor of a presidential republic, and expresses a strong sense of anti-Americanism and anti-Zionism. It is the only recognised party in Italy with the inscription "Fascismo" on its logo. Overall, it is an ultranationalist party with tendencies towards the Third Position and anti-capitalism.

== Controversies == Controversies and disputes have arisen with groups such as People for the Ethical Treatment of Animals (PETA), governmental and social agencies, and unions and trade groups over various topics. These situations have touched on legal and moral concepts such as animal rights, corporate responsibility, ethics, and social justice. In many of the cases, the situations raised legal questions, dealt with legal compliance, or resulted in legal remedies such as changes in contractual procedure or binding agreements between parties. The resolutions to these legal matters have often altered the way the company interacts and negotiates contracts with its suppliers and franchisees, or how it does business with the public. == Prognosis == Symptoms of fibromyalgia are regarded as persistent in nearly all patients. The German Federal Ministry of Health guidance on FM refers to a longitudinal study (n = 1555) in which 10% of patients had substantial improvement in pain, 15% had moderate improvement, and 39% worsened. FM severity worsened in 36%. No average meaningful change in symptoms was found, and high levels of self-reported symptoms and distress continued for most patients. Fibromyalgia can cause substantial disability, with impacts on the ability to work, engage in recreational activities, and do household duties. Amongst people with fibromyalgia, the proportion that are able to maintain their jobs has been estimated at between 34% and 77%, and some research suggest the typical reduction in work hours is between 50-70%. There is variation in the level of impact, with 71.4% of those who are mild remaining in employment, compared to 61.2% of those who are moderate and 28.5% of those who are severe. Levels of pain can vary significantly over time, from severe pain to almost symptom-free phases. A 2023 meta-analysis found that FM people were at a standardized mortality ratio (i.e., observed mortality rates in the study population, compared to expected levels based on a standard population) of 3.37 (95% CI 1.52 to 7.50) for mortality due to suicide.

== Research == There are various studies researching the concept of the wounded healer, most notably that by British counselor and psychotherapist Alison Barr who studied the significance of psychological wounds on people who decide to train as counsellors or psychotherapists. Barr used a pluralistic approach to her research, with the quantitative data analyzed using descriptive and inferential statistics and the qualitative data analyzed using thematic analysis, with a grounded theory approach. An on-line questionnaire was conducted with 253 respondents. Pilot and verification studies were performed, and opportunities for further research highlighted.
